The BCA Protein Assay Kit is one of the most commonly used protein concentration assays. BCA protein quantification involves a two-step reaction: firstly, divalent copper ions (Cu²⁺) are reduced to monovalent copper ions (Cu⁺) by the peptide bonds of the protein under alkaline conditions; secondly, two molecules of BCA complex a monovalent copper ion (Cu⁺) to form a purple complex with a strong absorption value at 562nm, and the absorption value of the complex correlates linearly with the concentration of the protein within a certain range.
