Fluorescein isothiocyanate (FITC) labeling uses pre-activated fluorescein reagents to react with the primary amine groups in proteins, thereby attaching fluorescein to the protein molecules. The maximum excitation wavelength (Ex) of fluorescein is 490-495 nm, and the maximum emission wavelength (Em) is 520-530 nm, resulting in a bright yellow-green fluorescence.
