Initially, the cell pellet must be lysed using methods such as freeze-thaw lysis, alkaline lysis, or ultrasonic fragmentation. Following 6FF chromatography, most RNA is removed. High-purity covalently closed circular DNA (cccDNA) is then obtained using Smac PlasmidSelect affinity resin. Finally, strong anion exchange chromatography is used to reduce endotoxin levels, ensuring the process meets drug registration standards.
