PreScission Protease (PPase) is a prokaryotic expression product of Human Rhinovirus 3C Protease. It is able to specifically recognizes the short peptide Leu-Phe-Gln/Gly-Pro and cleaves between Gln and Gly residues. PreScission Protease recognizes the enzyme recognition sequence (LFQ/GP) carried in the vectors PGEX-6P-1, PGEX-6P-2, and PGEX-6P-3, which can cleave the GST tag from the fused target protein. The sequence (LFQ/GP) can also be accessed into other vectors by cloning. During the cleavage reaction, it is suggested to find out the best conditions for different cleavage times and evaluate the degree of cleavage by SDS-PAGE. The amount of complete digestion, temperature and incubation time of GST fusion protein precisision protease should be changed according to different target proteins, and the cutting conditions should be optimized during the experiment. It has a GST tag at the N-terminal which can be conveniently removed after digestion by Glutathione Beads (Smart-Lifesciences, Cat. No. SA008010). This product is not supplied with a buffer for digestion.
