The Smart Assembly Cloning Kit is a simple, fast and highly efficient DNA cloning kit that enables targeted cloning of inserts into any site of any vector. The vector is linearized in an arbitrary manner by introducing the terminal sequence of the linearized vector at the 5' end of the insert's forward/reverse PCR primer, resulting in PCR products with sequences at the 5' and 3' ends that are identical to those at the ends of the linearized vector (25 bp - 40 bp). The PCR product with the terminal sequences of the vector and the linearized vector are mixed in certain ratios and reacted at 50℃ for 15 min with recombinase to achieve targeted cloning. As a new generation of recombinant cloning kits, its core advantage over similar products is that it is optimized for multifragment recombination reactions, allowing sequential splicing of up to 12 inserts at a time.
