Streptavidin is a 66 kDa tetrameric protein. Each mole of streptavidin binds to four moles of biotin, which can be used to detect and isolate biotinylated proteins. Horseradish peroxidase (HRP) couples streptavidin by cross-linking the amino group on streptavidin to the carbohydrate group on HRP. Streptavidin-HRP binds to biotinylated proteins and specific chemiluminescent substrates to generate light signals. Streptavidin-HRP coupling has a very high conversion rate and can produce high sensitivity in a short reaction time.
