Streptavidin is a biotin-binding protein found in the medium of Streptomyces avidinii. Each mole of streptavidin binds to four moles of biotin, and the high affinity between the two is almost unmatched in natural products (dissociation constant about 10-15 mol/L). Streptavidin has no carbohydrate side chains like avidin and has a near-neutral isoelectric point that facilitates the conduct of most useful biological reactions (streptavidin has a pI value of 5-6 compared to 10 for avidin). Therefore, streptavidin tends to exhibit lower levels of nonspecific binding than avidin.
