Sulfo-SMCC is a class of water-soluble bifunctional coupling agents containing N-hydroxysuccinimide (NHS) and maleimide groups that bond to compounds  containing amine and sulfhydryl groups, respectively. NHS reactive esters form amide bonds with primary amines at pH 7-9, while maleimides form stable thioether bonds with sulfhydryl groups at pH 6.5-7.5.

Sulfo-SMCC is commonly used in the preparation of antibody-enzyme couplings or semi-antigens. Generally, a two-step process is recommended. In the first step, the excess Sulfo-SMCC is reacted with amino-containing proteins, and the unreacted Sulfo-SMCC is removed by desalting columns or dialysis. In the second step, the sulfhydryl-containing proteins are linked to the modified amino-containing proteins.
