TEV Protease is a recombinant protease extracted from Tobacco etching virus (TEV), which is modified to be more stable than the natural TEV Protease and is often used to remove the affinity tag of purified fusion proteins. TEV Protease has highly site-specific, recognizing the seven amino acid sequence of EXXYXQ (G/S), most commonly ENLYFQG, whose cleavage site is located between glutamine and glycine or serine. The protease cut multiple amino acid sequences at the G/S (or P1) site, providing the required N-terminal amino acid for the cleaved fusion portion. TEV Protease achieves optimal activity at pH 7.0, 30°C, but actives over a wide range of pH 6.0-8.5 and 4-30°C, allowing modification according to target protein selection reaction conditions. It has a His tag at the N-terminal which can be conveniently removed after digestion by Ni NTA Beads(Smart-Lifesciences,Cat. No. SA004010). This product is not supplied with a buffer for digestion.
