Recombinant Protein/VLP Vaccines
07/05/2024The preparation process of recombinant protein/VLP vaccine is similar to that of general recombinant protein preparation. However, since the expressed protein is generally a recombinant surface antigen, VLP (eg: 9-valent HPV vaccine) or some kind of polymer (eg: RSV vaccine) will be automatically formed in most cases. In the final stage of purification, attention should be paid to removing monomers and poorly formed VLPs.
It is worth mentioning that since some VLPs have large particle sizes, they are more likely to encapsulate impurities during the formation process, and such VLP particles will cause side effects to the human body. Therefore, it is necessary to inhibit VLP generation in the early stage of purification, and promote VLP generation by changing the liquid when the purity is high, or remove the VLP particles formed in the early stage.
It should be noted that ion exchange chromatography may destroy the VLP structure during adsorption, causing VLP disintegration. Therefore, attention should be paid to the surface charge effect when designing the antigen structure.
